Almost 5,000 fans packed into London's Copper Box to cheer on Joe Prunty's side, and they rose to the occasion - with Dan Clark the top scorer on court with 23 points.
Nine spaces separate these sides in the world rankings, but Britain bridged that gap impressively with Clark at the top of his game.
The power forward scored nine points in the opening quarter, which ended 22-22, and remained a constant threat along with Gabe Olaseni and Luke Nelson.
The match remained tight but Greece kept their noses ahead throughout to clinch victory.
"It was a great game. We've had issues with turning the ball over but we've cleaned that up," Prunty said.
"Defensively, some of the things they got were too easy in the paint and we had a few defensive breakdowns but overall it was a positive game, well fought and we've just got to learn from the mistakes we made."
Britain have been drawn in a tough Group D for the European Championships, along with the top-ten ranked nations Serbia, Turkey and Russia.
