Humphries and brakeman Melissa Lotholz clocked a two-run time of 1:45.37, beating the Whistler Sliding Centre track record on both occasions.
The previous record was 52.85 seconds, set by Humphries on her way to gold at the 2010 Olympics, but now stands at 52.66.
America's Jamie Greubel Poser finished second in 1:45.53 while Austrian Christina Hengster completed the podium with a time of 1:45.64.
"There was a lot of pressure, being the Olympic champion here and being at home," said Humphries.
"I don't like the pressure, but I have experience and know how to deal with it. It is one of the things that comes with competing in sport at this level.
"This is what we compete for, what we love and it is high-performance sport at its finest."
That was followed by the two-man bobsleigh, which saw Canada's Chris Spring claim his first ever World Cup gold.
Alongside Lascelles Brown, the 31-year-old from Calgary set a two-run time of 1:42.76 to ensure Latvia's Ugis Zalims could only clinch silver and Russian Alexander Kasjanov had to settle for bronze.
"There's no better feeling than being a winner on your own track," said Spring.
Humphries now holds a 50-point lead at the top of the World Cup standings, while Spring's victory moves him up to 11th. South Korea's Yunjong Won, who finished ninth in Whistler, stays top.
