The first of six events, 15-year-old Medvedeva was in impressive form at Progressive Skate America, scoring 70.92 for her program.
American Gracie Gold currently lies second after a score of 65.39 while Satoko Miyahara of Japan is third, but there was no doubting who the star of the day was.
"I am overall quite pleased," world junior champion Medvedeva said. "There was a little mistake in the combination, but I don't think it affected my performance.
"It's the second season that I have been using this layout of my program (putting all jump elements into the second half). At the beginning, it was a little harder, but now I am used to it and obviously I'm doing it to get more points."
First on the ice were the pairs and Americans Alexa Scimeca and Chris Knierim gave the home fans plenty to cheer early on as they too picked up a personal best score of 69.69 to take the lead.
They are followed closely by Chinese duos Wenjing Sui and Cong Han and Xuehan Wang and Lei Wang who sit second and third respectively.
There was more for the crowd to shout about as Madison Chock and Evan Bates took the lead in the ice dance with a score of 70.56.
Russians Victoria Sinitsina and Nikita Katsalapov are second on 62.76 while Canadians Piper Gilles and Paul Poirer are third on 61.33.
And in the men's competition Max Aaron scored a personal best 86.67 to narrowly lead Han Yan of China by just 0.14 after the short program.
