With Barceloina leading 7-0 from the first leg of the semi-final at the Camp Nou, Messi's absence for the trip to Mestalla is hardly going to hurt Luis Enrique's side.
According to the Spanish newspaper AS, the Argentina forward "left hospital after undergoing lithotripsy shock wave treatment, a non-invasive procedure to break up kidney stones, on Tuesday".
Barcelona have confirmed that Messi is due to return to training on Wednesday with Enrique likely to make full use of the Barca squad holding such a heavy lead from the first leg against Gary Neville's side.
Messi missed the semi-final of the Club World Cup against Guangzhou Evergrande in December due to a kidney problem.
He is likely to return for Barcelona's next Liga match with Celta Vigo at the Camp Nou on Sunday evening.
Barcelona are three points clear of Atletico Madrid at the top of the standings with one game in hand.
