The 24-year-old centre-back, who moved to the Bernabeu from Lens in 2011, has helped Real win La Liga once and the Champions League three times.
A statement published on Real's official website said Varane's new deal would run until the end of June 2022.
"Real Madrid and Varane have agreed to extend the player's contract, which will see him remain at the club until 30 June 2022.
"Tomorrow, Thursday, at 2pm (1pm UK time), Varane will appear before the media in the press room of the Santiago Bernabeu stadium."
The player was still buzzing following Real's 3-1 victory over Borussia Dortmund in their Champions League clash on Tuesday evening.

There will be a number of top clubs across Europe who are very disappointed as a result of this news with Jose Mourinho having been the most consistently linked with a move for his former player. It was no secret that Manchester United were keen to add Varane to their ranks but Real have again moved to keep one of their best players.
Zinedine Zidane will be delighted that Varane is now contracted to remain at the club until 2022 and, at the age of 24, should have his best years ahead of him through this deal. Varane played superbly again against Dortmund in what was a very impressive win for the defending champions and his influence continues to grow at the club.
