Mustafi, 24, is the second signing of the day for Arsenal with Spanish forward Lucas Perez joining from Deportivo for £17.1m. - making Tuesday's £52.1m the most Arsene Wenger has ever spent in one day.
Wenger said of his new defensive recruit:
He is at the right age. He has good experience. He is a very focused player who can play with the ball as well. We have taken a great player but have prepared well for the future.
Mustafi made 64 Liga appearances over two seasons with Valencia and has Premier League experience having come through the Everton academy.
He arrives in London as a competitor to former international team-mate Per Mertesacker, and summer signing Rob Holding, in the tussle to partner Laurent Koscielny at the heart of the Arsenal defence.
Arsenal are also clearing their squad out with Calum Chambers joining Middlesbrough on loan on Tuesday night.
Jack Wilshere has also been given permission to find a club on loan before the close of the window on Wednesday night at 11pm, and winger Serge Gnabry is set to join Werder Bremen on a permanent deal.
