Marcelino Garcia Toral's side continued their superb start to the season with a convincing victory over the Andalusians, who offered little resistance at a vibrant Mestalla.
Guedes followed up his brilliant strike against Real Betis last weekend with another stunning goal, beating two men and then rocketing a shot past the helpless Sergio Rico in the 43rd minute.
Simone Zaza doubled Valencia's lead six minutes after the restart, beating defender Simon Kjaer and clinically striking a shot into the far corner for his eight goal of the season.
Santi Mina tucked home the third late on after being played in by the electric Guedes, with the 20-year-old Portuguese winger grabbing his second in stoppage time, as unbeaten Valencia moved to 21 points from nine games, staying second.
Barcelona host bottom side Malaga later on Saturday, while Levante drew 1-1 with Getafe and Real Betis beat Alaves 2-0 earlier on in the day.
