The DS Virgin Racing driver leapt to the top of the times midway through FP2, deposing Saturday's race winner Felix Rosenqvist in the process.
Rosenqvist set the early pace on a 1m07.753s, which Lopez edged by almost a tenth to set a new Tempelhof record of 1m07.670s.
Lopez's effort meant Sebastien Buemi's hold of the lap record lasted little more than an hour - the Renault e.dams driver having topped FP1 with a 1m07.712s.
Buemi was in the clear in the opening session, opting for full-power 200kW laps while the other frontrunners focused on race-power runs.
In FP2 he left his qualifying simulation until the last moment, then jumped to third with a 1m07.782s.
That dropped the second DS Virgin of Sam Bird to fourth ahead of Daniel Abt.
A lurid Jean-Eric Vergne ended up sixth-fastest, having lost time with a sideways moment through the final corner.
Lucas di Grassi lapped faster than his Saturday pole time but was still only ninth fastest, half a second off the pace, as a host of drivers improved in warm conditions and with the track at its most rubbered-in.
