The five-times world champion will meet Judd Trump or Mark King in the quarter-finals at Venue Cymru in Llandudno.
The Englishman pieced together breaks of 72, 65, 118, 101 and 71 in one of his best performances of the season.
English Open champion Wenbo missed a black to beat O'Sullivan in the first round of the Masters in January before suffering a 6-5 defeat, but was never really within touching distance of his close friend.
"It's a nice event, but I've got to get out of Llandudno for a few days. It's a bit windy, there's not much to do around here," said O'Sullivan, who is due back in action on Wednesday evening.
There was also a comfortable win for Anthony Hamilton - recent German Masters champion - who completed a 5-1 victory over Stuart Bingham.
Hamilton rolled in runs of 131, 105 and 59 to secure a meeting in the last eight with Ding Junhui or John Higgins.
