Murphy’s run of 89, 101, 101 and 104 in just five frames saw him book a clash with Gary Wilson, a 5-3 victor over Graeme Dott.
Stuart Bingham struck a match-high break of 96 as he beat Scott Donaldson 5-3.
Bingham reeled off the opening three frames to put himself in a commanding position before addressing a mini wobble with another half century in the final frame.
Jimmy White also progressed with a 5-1 win over wild card Li Yuan.
Meanwhile, Mark Selby recovered from a slow start to power into the first round - with his qualifying match held over to the venue stage.
The world number one struggled in the early stages of his match against Adam Stefanow, going 2-0 down before getting on the board with a break of 61.
World champion Selby levelled the contest with a 136 and, although his Polish opponent regained the lead by winning the fifth frame, knocks of 113 and 126 put Selby on the brink of victory and he wrapped up the success with an 87.
Judd Trump had no problems as he started his title defence with a 5-0 whitewashing of Jason Weston 5-0, also reaching the first round proper.
