The welterweight - who remains one of Britain's highest-profile fighters despite 18 months of inactivity - and Matchroom will officially announce a multi-fight deal at a press conference in London on Wednesday.
Press Association Sport understands that before signing with Matchroom, Khan rejected an offer from ITV, having recently appeared on their programme I'm A Celebrity... Get Me Out Of Here.
By agreeing terms with Hearn he will instead work with a promoter he has regularly spoken of his dislike for.
Hearn has long annoyed Khan by attempting to lure him into what would be a significant domestic fight with another of his fighters, and the 31-year-old's long-term rival, Kell Brook. He has also accused him of pricing himself out of what would prove a highly popular match-up.
When discussing the promoter in August from Las Vegas during the build-up to Floyd Mayweather's fight with Conor McGregor, Khan said: "The only way I would take that Brook fight is if he left Eddie Hearn. I just don't like Eddie Hearn.
"He's very disrespectful and he's never come clear with me. If he really wanted the fight between me and Kell Brook he should not be speaking to me, he should be speaking to (my adviser) Al Haymon. I always said that to him.
"That's one of the reasons why I keep away from Eddie Hearn and his shows, I just don't want to be in that drama."
While their agreement will increase expectations of an eventual fight with Brook, that his rival has since stepped up to the light-middleweight division from 147lbs means it is far from guaranteed.
In addition to joining the same stable and television network as Brook, Khan will also be joining that of world heavyweight champion Anthony Joshua, who he last year wrongly accused of having a relationship with his wife Faryal Makhdoom.
The unexpected agreement regardless means that after his stoppage defeat by Saul 'Canelo' Alvarez in May 2016, his troublesome hand injury and issues in his personal life since then, Khan will finally resume his career, as he has long wished to do.
