The defending champion has 118 points after a near-flawless opening day, with Belgium’s Jolien D’Hoore on 110 and the United States’ Sarah Hammer on 108.
Trott, who became the first British woman to earn three gold medals, is targeting a fourth Olympic triumph following her London 2012 double and last week’s victory in the team pursuit.
She opened her quest with second in the scratch race before storming to victory in the individual pursuit and elimination race.
The time trial, flying lap and points race await on Tuesday.
