Welbeck went down under pressure from Milan defender Ricardo Rodriguez but replays suggested it was a soft decision.
The England forward converted the penalty to level the match on the night as Arsenal came from a goal down to triumph 3-1.
Speaking after the match Wenger told BT Sport that he had not seen the incident.
"Honestly I don't know. When I watch the penalty, I will give you an honest answer," said Wenger.
"It was good to see a response from the team at 1-0 down.
I don't want to accuse Danny Welbeck of diving before I see it again.
Wenger, whose side have now won three in a row in all competitions, was also pleased with his team's turnaround after a tricky period.
The Frenchman added that he would like to see his side avoid favourites Atletico Madrid in the quarter-final draw which will take place on Friday.
"It's much better, we needed that to recover from our results," Wenger added.

"It shows how the team has responded. We had a difficult game but in the end we won it.
It was an open game. We played backwards a bit too much in the first half to protect the result. We played a very good side. We have to improve defensively, but overall we are team with technical quality which is more offensive-minded.
"We struggle a bit more when we have to defend. We finished the game more compact and more solid. We would certainly want to avoid Atletico Madrid, but we will see, we have no influence over that."
