Defoe has struggled for form and game time since moving to the Cherries from Southampton on a free transfer over the summer.
He has scored just once with Callum Wilson (4) and Joshua King (3) leading Bournemouth's scoring charts.
Southampton return to a back four after playing with a back five in mid-week against Manchester City with Maya Yoshida dropping out.
Cedric Soares, Pierre-Emile Højbjerg, Mario Lemina, Nathan Redmond and Shane Long also miss out as Jeremy Pied, Steven Davis, James Ward-Prowse, Charlie Austin, Tadic and Boufal all come in.
A win for the Saints will see them move up to tenth, ahead of Everton and Brighton.
If Bournemouth win they will overtake today's opponents and move up to 11th.
Bournemouth - Begovic, Adam Smith, Francis, Ake, Daniels, Stanislas, Lewis Cook, Surman, Fraser, King, Defoe
