Lamela, 26, signed for Spurs from Roma in 2013 for a then club-record fee of £30 million.
He has made 149 appearances for the club in all competitions although his progress has recently been hampered by a hip injury.
During his time out injured manager Mauricio Pochettino regularly bemoaned his absence, with Lamela a key part of the high-intensity system he likes to play.
The Argentine returned to action this season and scored twice in the 5-4 victory over Leicester City on the last day of the season.
Lamela's new contract will run for four years and the news comes shortly after team-mate Son Heung-min signed a new until 2023.
The pair follow Pochettino and Harry Kane in signing new contracts with reports that more first-team players are expected to follow suit.
