Kane, 25, suffered the injury during Sunday's 1-0 loss to Manchester United at Wembley and he was seen hobbling off the pitch after the full-time whistle
Initially the swelling meant that Spurs could not do a full evaluation of the injury but they have now released a statement confirming it is ankle ligament damage.
"Following preliminary assessments, we can confirm that Harry Kane has damaged ligaments in his left ankle, sustained during Sunday's match.
"He will continue to be monitored by our medical staff as he commences rehabilitation and is expected to return to training in early March."
It is the fourth year in a row that Kane has suffered ankle problems and it comes as the in-form Son Heung-min has left to join up with South Korea's squad at the Asian Cup.
If Kane does not return until March he will miss the second leg of the Carabao Cup semi-final against Chelsea at Stamford Bridge on January 24 as well as the Premier League clash at the same venue on February 27.
He will also miss the first leg of the Champions League last 16 tie with Borussia Dortmund at Wembley on February 16.
The start of March sees two pivotal matches for Spurs as they face Arsenal in the North London derby on the 2nd and the second leg against Dortmund on the 5th.
It leaves Mauricio Pochettino with just Lucas Moura and Fernando Llorente to call upon, with the latter linked heavily with a return to former side Athletic Club.
Kane posted a message to his social media channels on Tuesday evening thanks fans for their messages.
Kane wrote "Injuries are part of the game but no-one will be working harder to get back fit. Thanks for the messages."
