Leicester raced into an early lead and were 28-9 up at half time, tries coming from Tom Varndell, Ben Kay, Tom Croft and Alesana Tuilagi.
James Hook's three penalties kept Ospreys barely in touch following an error-ridden first 40 minutes that gifted the Tigers their scores, scrum-half Justin Marshall in particular having a poor game.
Lee Byrne opened the second half scoring when he crossed for Ospreys four minutes in, and two tries followed from Shane Williams to make it 31-28 to Leicester after an hour.
Varndell then grabbed his second try of the game, and Andy Goode added a penalty to extend Leicester's lead to 41-28.
Nikki Walker scored for Ospreys with six minutes left on the clock, and Hook's conversion ensured a tense final run-in for both sets of fans.
Leicester held on though to complete the first stage of their potential treble - the Heineken Cup and Premiership title still well in their sights.
