Quins announced that their 28-year-old captain had committed his "long-term future" to the club.
Care has won 53 caps and made more than 180 appearances for Quins since joining them in 2006.
"The re-signing process was done very quickly as there was never any doubt in my mind to go anywhere else," Care said.
"I love being here, I am part of a great group of people and I'm really excited to have pledged my commitment to the club for the next few years."
Care has scored more than 50 tries in Quins colours and played an integral part during the club's Premiership title-winning campaign in 2012.
Quins rugby director Conor O'Shea added: "Danny is the ultimate game-changer and has matured and grown in front of us all.
"In Danny, we see the Harlequins style stamped all over the way he approaches the game.
"Captaincy is another natural progression for him, and I am looking forward to watching him kick on and become an even better player.
"In my opinion, with the skill-set he possesses, he should go on to win 100 caps for England and win more silverware for the club."
