Fresh from winning his fourth ranking event of the season at the World Grand Prix on Sunday, world number two O'Sullivan made light work of 2006 world champion Dott
A superb clearance in the first frame enough to win it on the black after recent German Masters finalist Dott had produced a fine 64.
Runs of 106 and 69 gave O'Sullivan a 3-0 lead despite his opponent opening with a knock of 50 at the Motorpoint Arena in Cardiff.
It was clear the five-times world champion had the match won as a deflated Dott could not capitalise on a rare scoring chance in losing the fourth frame.
He will face David Grace for a place in the last 16.
Later, Neil Robertson was beaten 4-3 by world number 100 Ian Burns.
The 'Thunder from Down Under' looked set to book his spot in the next round after producing runs of 100, 100 and 130 to lead 3-2 in the best of seven second-round encounter.
In fact, such had been the Australian’s brilliance, he had a 100% pot success rate as the match entered its sixth frame.
However, Burns, who had produced breaks of 85 and 106 on his way to taking two of the first three frames, produced a fine 75 to square the match after Robertson missed his first pot of the day, a tough green to the middle.
And the 32-year-old from Preston held his nerve to see it out and progress to the next round.
Mark Williams beat Aditya Mehta 4-3 in a match most notable for its epic 73-minute third frame.
