The seventh seed from the Czech Republic needed two hours and 28 minutes to overcome Gasquet as he reached the quarter-finals of the Masters tournament for the seventh time in 12 appearances.
Berdych, 30, reached the final six years ago, but lost to retired American Andy Roddick.
His reward is a meeting with world number one Novak Djokovic or Dominic Thiem - the 14th seed - for a place in the last four.
Berdych was a break down in the opening set, but won four straight games from trailing 4-2 to pocket the set.
Gasquet broke early on his way to restoring parity at 1-1 before Berdych broke in the 11th game of the decider to swing the match in his favour.
He served out to love to progress.
In other matches, David Goffin reached his second successive Masters quarter-final with a 7-5 6-3 win over Horacio Zeballos.
He will meet Gilles Simon for a place in the last four having lost to Milos Raonic in the semi-finals of Indian Wells last week.
Simon needed only 58 minutes and conceded only 11 points on serve on completing a 6-0 6-1 win over Lucas Pouille.
