Seven-times champion Roger Federer was in imperious form as he demolished Frenchman Gilles Simon 6-3 7-5 6-2 on Court One to reach Friday's semi-finals.
Federer leads the head-to-heads 12-11, but Murray won the last of their duels on grass when the Scotsman picked up gold for GB in straight sets at the 2012 London Olympics.
Federer had left Murray in tears only a few weeks earlier by downing the British number one in the Wimbledon final, and he has won the past three matches between the men, the last being a 6-0 6-1 drubbing of Murray at last year's World Tour finals indoors in London.
It is Murray's sixth Wimbledon semi-final, and he said: "I love playing here, and I've always played some of my best tennis here. I hope I can get to another final this year, but is is going to be tough obviously.
"During the matches, there is quite a lot of pressure...I know Roger very well. We've played each other many times. We get on well. We've played some good ones (matches) in the past."
Murray was watched by Prince William, his wife the Duchess of Cambridge and former England football captain David Beckham.
He broke serve for the first time in the third game of the match to lead 3-1 when light rain forced play to be suspended.
He proceeded to see out the set only for another rain delay to force the Centre Court roof to be shut with Murray trailing 4-3.
Pospisil continued to look uncertain under the roof, and dropped his serve at 5-5 in losing the second set.
He earned his first break point of the match with Murray a break up and 5-4 ahead in the third set, but could not convert as the physical demands of going 10 sets in his previous two matches appeared to finally catch up with him.
